Conditional expression of the human C-terminal-truncated (1-119) mutant (A53T) alpha-synuclein under the control of the endogenous ROSA 26 promoter in a Cre recombinase -dependent manner. Breeding with TH-Cre transgenic mice allows the selective expression of the transgene in dopaminergic neurons.
Endogenous mouse alpha synuclein: Yes.
Corresponding human genotype: The A53T mutation causes a rare autosomal dominant familial form of PD. The truncated form of alpha-synuclein detected in brain tissue from post mortem PD brain and enriched in Lewy bodies

Transgene expression
- 10 months: Transgene expression is observed in TH positive neurons and/or terminals, including olfactory bulb, striatum, cerebral cortex and ventral midbrain. A low expression level of the mutant protein is observed compared to that of endogenous mouse alpha-synuclein
Neurodegeneration
- 10-12 months: No loss of TH-positive neurons is observed.
No change in survival observed up to 18 months of age.
